B
Grade Bprobable, not settledPhase 3 sub-analyses · effect size: large
Short answerProbable, not settled. Human data exist (Phase 3 sub-analyses) and mostly agree, showing a large effect — but too few, too small, or not yet peer-reviewed for a firm answer. Among participants with baseline liver fat above 10%: −66% (4 mg) and −80% (6 mg) versus −5% in GLORY-1; −72% versus +5% in GLORY-2.

Among participants with baseline liver fat above 10%: −66% (4 mg) and −80% (6 mg) versus −5% in GLORY-1; −72% versus +5% in GLORY-2.

Human trials exist and mostly agree, but there are too few of them, they are too small, or one big one is still reading out. Expect the grade to move.

In context

Mazdutide is a once-weekly GLP-1/glucagon dual agonist developed by Innovent with Lilly. In the 610-person GLORY-1 trial, published in the New England Journal of Medicine in May 2025, it produced 14.8% weight loss at 48 weeks on 6 mg against 0.5% on placebo; a 9 mg dose reached 18.6% at 60 weeks in GLORY-2. China's regulator approved it for chronic weight management on 27 June 2025 and later for type 2 diabetes. It is not approved in the US, EU or UK.

Mazdutide is approved in china only. Legal status differs by country — see the regulation section or the jurisdiction pages.
